My research is divided into semisynthesis and precursor-directed biosynthesis (PDB) of polyketides featuring antibacterial activities.
The PDB approach exploits the promiscuous polyketide synthase (PKS) governing the biosynthesis of erythromycin in Saccharopolyspora erythraea. Macrolides are also semisynthetically derivatized to explore structure-activity relationships in close collaboration with crystallographers and molecular modelling groups (see research and collaborations). Regio- and chemoselective synthetic modifications give rise to new compounds with the aim of overcoming resistances and improving the antibacterial activity.
